This lot is a result of two subcultures made in L1 medium at 20º C. Approx.10mL of overlying medium was removed from each tube, the remaining biomass combined, and then 12 mL of that was mixed with 3 mL of 30% CPA. 1mL of that was dispensed into each cryovial. The vials were exposed to cryoprotectant for 35 minutes before placing in a control rate freezer . Three test vials were also frozen.
Three test vials were thawed at 20º C until there was just a little ice left. They were then spun at 7000 rpm for approx. 10 seconds in an eppendorf centrifuge to pellet the cells. The supernantant was removed and the cells resuspended and pipetted into room temperature tubes of L1. They were grown at 20º C in high light. The thaws were shippable after 30 days. The bacto tests showed that the thaws were contaminated with bacteria as in the active collection. No fungal contamination was detected.
