This lot is a result of two tubes sub-cultured in L1 medium at 14º C. About 8mL of overlying medium was removed from each tube. The remaining culture from one tube was combined was the other, then mixed. 10.5mL of that mixture was added to 4.5 mL of 30% CPA, and mixed again. 1mL of that was dispensed into each cryovial. The vials were exposed to cryoprotectant for 29 minutes before placing in the control rate freezer. Three test vials were also frozen.
Three test vials were thawed at 20º C until there was just a little ice left. They were then spun at 7000 rpm for approx. 10 seconds in an eppendorf centrifuge to pellet the cells. The supernantant was removed and the cells resuspended and pipetted into 14C temperature tubes of L1. They were grown at 14º C in high light. The control shippable after 14 days. The thaws were shippable after 28 days.The bacto tests showed that the thaws were contaminated with bacteria as in the active collection. No fungal contamination was detected.
