This lot is a result of a subculture made in 35% medium at 14º C. Approx. 5mL of overlying medium was removed, the remaining culture mixed, and then 9mL of that was mixed with 6mL of 30% CPA.  1mL of that was dispensed into each cryovial. The vials were exposed to cryoprotectant for 27 minutes before the fullsw program was started. The probe was in a final volume of 1ml and consisted of 10% DMSO diluted in 35%. Three test vials were also frozen.

Three test vials were thawed at 20º C until there was just a little ice left. They were then spun at 7000 rpm for approx. 10 seconds in an eppendorf centrifuge to pellet the cells. The supernantant was removed and the cells resuspended and pipetted into room temperature tubes of 35%. They were grown at 14º C in high light.  The thaws were growing and shippable after 28 days. The bacto tests showed that the thaws were contaminated with bacteria as in the active collection. No fungal contamination was detected.
