Q+A:

Q: I used L1 medium as a growth media and diluted the sample into 1:5 ratio. The sample is incubated at 22 oC. I checked the optical density (OD 600) of the sample with 3-4 days intervals. OD of stock solution (the sample sent by NCMA) had 0.10. The diluted sample had the OD of 0.02 and the OD of the sample didn't increase more than 0.03 over a month period. I was wondering whether there was something that I did wrong or the sample that I received was not a good one.

A: CCMP358 is on a one month transfer cycle. Every month a new child set is created with a 2 drop inoculate from the bottom of the vessel so the growth is pretty slow on this culture. All our cultures are grown in 18mL of medium and after they are inoculated there is no manipulation of the vessel. We don't use shakers or vortex the cultures at any time. Most of our cultures are at ~80-100uE.
