This lot is a result of two subcultures made 5/16/18 in DYV medium at 20º C. Approx.7mL of overlying medium was removed from each tube, the remaining biomass combined, and then 12 mL of that was mixed with 3 mL of 30% CPA. 1mL of that was dispensed into each cryovial. The vials were exposed to cryoprotectant for 32 minutes before placing in a minus 80 degreeC freezer . They were placed in an one inch thick Styrofoam box, of sufficient length to accommodate canes, to moderate the rate of cooling. This was done at about 8:15 am. The canes were moved to the LN2 storage tank that afternoon at around noon Three test vials were also frozen.

Three test vials were thawed on 5/17/18at 20ºC until there was just a little ice left. They were then spun at 7000 rpm for approx. 10 seconds in an eppendorf centrifuge to pellet the cells. There were no visible pellets so the entire contents of the vials were harvested and pipetted into room temperature tubes of DYV. They were grown at 20ºC in high light. The control was shippable after 35 days, the thaws after 35. The bacto tests showed that the thaws were axenic as in the active collection. No fungal contamination was detected.
