This strain was grown in 1.5L of 12ppt SW +768 at 22 degrees. When the culture was fairly dense (~480 cells/ml) the culture was vacuum filtered through 3um PC filters at <250 mm Hg vacuum. When the fliteration slowed, the concentrated culture above the filter and the filter itself was placed in a 15ml centrifuge tube. This process was repeated until the culture was concentrated down to approx. 15 ml. The resulting cell density that was dispensed into cryovials was ~11,400 cells/ml. The cells were exposed to the cryoprotectant for approx 20 minutes before the fullsw program was started. The probe was in the same final volume as the cells (1ml) and consisted of 10% DMSO diluted in f/2-si. Three test vials were also frozen.
The three test vials were thawed at 36 degrees into plastic vials with 15mL 12ppt SW + 1 drop dense CCMP768 culture. They were grown at 22 degrees under medium light. The bact test showed the thaws were contaminated with bacteria, as in the active collection. No fungus was detected.
