Strain Specific FAQs:
Q: After about 1 week when I checked the cultures I noticed many actively lysing cells but still some ok cells that were alive but not swimming. Since then I have been checking on the culture under the microscope about every 3 days and I keep finding more and more lysed cells and only 1 living cell on average per culture. The cultures are being kept at 15 C with a 12:12light:dark cycle at ~40 uEinsteins. Do you have any advice on how to help these cultures thrive and do you have any idea why the cells might be lysing?
Q: Are you noticing any precipitate in the L1 medium? CCMP304 will survive in L1+Si, although we grow it in L1- to reduce the chances of silica piercing the cells. At 15C you're at the top of the temp range, do you have a cooler incubator?
A: We did have a little precipitate in the last batch of L1+Si media we made. I will try to make a new batch of L1-Si up and transfer to see if that helps some. I can also try lowering the incubator temperature a bit. I have other cultures in the incubator (CCMP 113, CCMP 688, and CCMP 686) but they should be alright if I drop the temperature to 13 or 14°C. Hopefully a combination of those two fixes should do the trick and save the culture so I don't have to order it again.
