Thalassiosira sp.  

ThereThis lot is a result of a subculture made in L1 medium at 2º C. Approx. 5mL of overlying medium was removed, the remaining culture was mixed, and 9mL of that was mixed with 6mL of 30% CPA. 1mL of that was dispensed into each cryovial. The vials were exposed to cryoprotectant for 30 minutes before placing in a minus 80 degree C freezer. They were placed in an one inch thick Styrofoam box, of sufficient length to accommodate canes, to moderate the rate of cooling. The canes were moved to the LN2 storage tank 4 hours. Three test vials were also frozen.

Three test vials were thawed at 20º C until there was just a little ice left. They were then spun at 7000 rpm for approx. 10 seconds in an eppendorf centrifuge to pellet the cells. The supernantant was removed and the cells resuspended and pipetted into room temperature tubes of L1. They were grown at 2º C in high light. The thaws were shippable after 30 days. The bacto tests showed that the thaws non axenic as in the collection. No fungal contamination was detected.
