This lot is a result of two tubes sub-cultured 23OCT2015 in 35% medium at 14º C. About 8mL of overlying medium was removed from each tube. The remaining culture from one tube was combined was the other, then mixed. 10.5mL of that mixture was added to 4.5 mL of 30% CPA, and mixed again. 1mL of that was dispensed into each cryovial. The vials were exposed to cryoprotectant for 29 minutes before placing in a minus 80 degreeC freezer . They were placed in a one inch thick Styrofoam box to moderate cooling- the box of sufficient length to accommodate canes. This was done at about 10:15am. The canes were moved to the LN2 storage tank ~ 2:20pm. Three test vials were also frozen.

Three test vials were thawed on 30Oct17 at 20ºC until there was just a little ice left. They were then spun at 7000 rpm for approx. 10 seconds in an eppendorf centrifuge to pellet the cells. There were no visible pellets so the entire contents of the vials were harvested and pipetted into 14C tubes of 35%. They were grown at 14ºC in high light. The thaws after 27. The bacto tests showed that the thaws were contaminated with the bacteria as in the active collection. No fungal contamination was detected.
