This culture was started in a tube of f/2-Si at 24 degrees C. Approximately 10mL of overlying medium was removed before mixing and dispensing the culture. The cells were exposed to the cryoprotectant for 42 minutes before the fullsw program was started. The probe was in the same final volume as the cells (1ml) and consisted of 5% DMSO diluted in f/2-Si.

Three test vials were thawed into room temperature 25ml tubes of f/2-Si medium . They were spun for 5 seconds at top speed in an eppendorf microfuge to remove the supernantant.They were grown in at 24 degrees in high light. In 12 days all three were shippable. The bact test showed that the thaws were contaminated with bacteria as in the active collection. No fungal contamination was detected.
